Dot blot
analysis of native and denatured AAV1-AAV9, AAVrh10, AAVrh74 and AAVDJ capsids
(5E+09 capsids per dot). Capsids were denatured at 95°C for 5 min in sample
buffer.
The nitrocellulose membrane was blocked with 5% dry milk in PBST (PBS + 0.1%
Tween 20) for 45 min at RT. The primary antibody anti-AAVrh10 (intact particle)
mouse monoclonal, PR10 (Cat. No. 690107) was diluted in blocking buffer
(antibody concentration 100 ng/ml) and incubated for 1 h at RT. The secondary
antibody goat anti-mouse IgG HRP was also diluted in blocking buffer (antibody
concentration 250 ng/ml) and incubated for 1 h at RT. The bands were visualized
by chemiluminescent detection using Pierce™ ECL Western Blotting Substrate.

Background
The anti-AAVrh10 is a mouse monoclonal antibody, specifically recognizing a conformational epitope only present on fully assembled and intact AAVrh10 and AAVrh74 capsids. It does not detect denatured AAV capsids. The anti-AAVrh10 can be used in versatile applications, such as tracking the localization of viral particles, studying protein-protein interactions, facilitating AAV particle purification, determining total AAV capsid titers or for neutralizing experiments.
