About the LNP FluoGreen Anionic Peptide Delivery Kit:

PROGENs FluoGreen Anionic Peptide Delivery LNPs come as a fluorescently-tagged, dryed lipid emulsion, easy to formulate by bench-top addition of ethanol. The fluorescence tag is attached to the sphingolipid contained in the formulation (Ex/Em = 495/503 nm).

The composition and molar ratios of PROGENs FluoGreen Anionic Peptide Delivery LNPs are optimized for associating and delivering anionic peptides (pH > pI) by rendering electrostatic interactions and can be used to efficiently deliver the peptide to specific cell lines and animal models at a recommended concentration of 4 mg/ml. 

The high biocompatibility of the FluoGreen Anionic Peptide Delivery LNPs enables an efficient internalization of peptides whilst yielding high cell viabilities. This kit facilitates efficient cellular delivery while preserving peptide integrity and activity, making it an ideal choice for innovative peptide-based treatments. Furthermore, its lipid nanoparticle formulation can be adapted to various administration routes, facilitating smooth transitions from in vitro to in vivo applications through the development of personalized solutions. It has shown efficacy in rodents and zebrafish models across various administration methods.

If necessary, the formulation can be concentrated by using an Amicon Ultra Centrifugal Filter, SpeedVac or Rotavap in mild conditions (avoid overpassing 35 °C or drying out the samples). Samples can be concentrated up to 4-fold its original volume (i.e., to a final volume 250 µL).

Our FluoGreen nanoparticles can be used for cell or organ analysis by flow cytometry or confocal microscopy. However, for whole-body imaging, PROGEN can provide customized reagents labeled with fluorophores such as Cy7.5. We also offer customized solutions to adjust fluorophore concentration as needed.
Currently, we offer the green fluorochrome. However, you can contact PROGEN for a customized formulation where we can incorporate alternative fluorochromes: Cy5, Cy7.5, and other fluorescent dyes.
Green fluorescence remains stable for up to 1 year at -20 ºC prior to formulation. After preparation, it is stable for at least 7 days (maximum tested time) when stored at 4 ºC, protected from light. In live cells, the signal can be tracked for at least 7 days.
No, it does not. The fluorescence arises from molecules that are covalently linked to the lipids, but have no intrinsic effect. The fluorescence does not interfere with the efficacy of the formulation or the activity of the associated drug/biomolecule.
This will depend on the specific properties of each molecule. Please refer to the specific protocol of each reagent. For in vivo delivery, formulation can be adapted under request to reach specific requirements attending to the dose and route of administration. For small drugs, it can range from 1% to 15% (55 μg - 0,83 mg) with respect to the total amount of lipids (5,5 mg) per vial. We recommend starting with 1%, and then increasing the concentration gradually. For proteins and peptides, it depends on the selected kit and on the specific molecule, but it is possible to load at least 150 ug of protein/peptide per vial.
We have tested in healthy mice to get an insight, but detailed biodistribution should be done with relevant models and with encapsulated drugs / molecules as this affects the biodistribution. We have conducted s.c. (subcutaneous) administration using one or more imaging modalities as we have optimized labeling (PET, SPECT and fluorescent tracking), i.v. (tail vein injection and retroorbital), i.a. (intra-articular), and i.p. (intraperitoneal).
Once formulated, we recommend using the formulation within 24-48 h, according to the provided protocol.
It is not necessary for initial in vitro testing. There is not a big increase in viscosity and the pH from the formulation is suitable for in vitro testing. It Is also possible to use buffers to formulate, as indicated in the kit protocol.
Filtration through 0,45 μm or 0,22 μm will work to eliminate free precipitated compound to obtain the percentage of free compound and then calculate the percentage of encapsulation through the subtraction of the total and free concentrations, in relation to the total concentration.
